Organ morphogenesis: A metalloprotease prepares the way

چکیده

It is generally believed that structural changes in the extracellular matrix are necessary for cell migration, wound repair and tissue remodeling. Matrix metalloproteases are the major family of proteolytic enzymes responsible for degrading extracellular matrix components, and have been implicated in all of these biological processes. They form part of a superfamily of zinc peptidases known as the metzincins, which include four distinct families: the astacins, the matrix metalloproteases, the adamlysins/reprolysins and the serralysins (large bacterial proteases). Members of this superfamily are characterized by both sequence and structural similarities, in particular an identical zincbinding environment [1].

Purification and Characterization of 50 kDa Extracellular Metalloprotease from Serratia sp. ZF03

Background: Proteolytic enzymes have an important role in variety of physiological and pathological functions. They have been used in therapeutic and pharmaceutical applications. Characterizations of extracellular proteases from various strains of S. marcescens indicate that most strains produce a very similar major metalloprotease. This metalloprotease (serrapeptidase, serrapeptase) is an impo...

Eversion and withdrawal of an intromittent organ before sexual maturation prepares male beetles for copulation

Some species of criocerine beetles have a hyper-elongated part of the intromittent organ called a flagellum. In resting position, the flagellum is stored in a specialized internal sac in the intromittent organ. This specialized state of the flagellum and internal sac is indispensable during copulation for flagellar insertion into the female spermathecal duct for sperm transfer. However, the mor...

The conserved oligomeric Golgi complex acts in organ morphogenesis via glycosylation of an ADAM protease in C. elegans.

In C. elegans, the gonad acquires two U-shaped arms through directed migration of gonadal distal tip cells (DTCs). A member of the ADAM (a disintegrin and metalloprotease) family, MIG-17, is secreted from muscle cells and localizes to the gonadal basement membrane where it functions in DTC migration. Mutations in cogc-3 and cogc-1 cause misdirected DTC migration similar to that seen in mig-17 m...

Notch modulates VEGF action in endothelial cells by inducing Matrix Metalloprotease activity

BACKGROUND In the vasculature, Notch signaling functions as a downstream effecter of Vascular Endothelial Growth Factor (VEGF) signaling. VEGF regulates sprouting angiogenesis in part by inducing and activating matrix metalloproteases (MMPs). This study sought to determine if VEGF regulation of MMPs was mediated via Notch signaling and to determine how Notch regulation of MMPs influenced endoth...

ADAM, a novel family of membrane proteins containing A Disintegrin And Metalloprotease domain: multipotential functions in cell-cell and cell- matrix interactions

C ELL--cell and cell-matrix adhesion, as well as proteolysis of the extracellular matrix, are vital for normal processes such as tissue morphogenesis and wound healing, as well as for pathologies such as tumor cell invasion and metastasis. A variety of cell surface adhesion proteins and proteases are important players in these events. Families of membrane-anchored cell surface adhesion molecule...
